Rapid Cell-Free Expression and Purification of B-lymphocyte Antigen CD19 Using eProtein Discovery™
In this Application Note, we leverage the eProtein Discovery™ system to screen 192 expression conditions and optimize the production of soluble CD19 protein within 24 hours. We successfully expressed and purified three CD19 length variants: full-length CD19, the N-terminal extracellular domain, and the C-terminal intracellular domain, overcoming challenges associated with "difficult-to-express" proteins.
From reading this, you'll gain an understanding of the following:
- Production of CD19 Variants: Evidence of the successful expression and purification of three CD19 length variants, including full-length CD19.
- Expression of Difficult Domains: How the eProtein Discovery™ system facilitated the expression of the challenging CD19 N-terminal extracellular domain.
- Scalability: The ability to scale up optimized expression conditions to produce microgram quantities of protein in 24 hours.
- A Reference for Other Proteins: A valuable reference for expressing other “difficult-to-express” proteins with transmembrane domains, disulfide bonds, and highly disordered regions.
